RESUMO

Objective: To identify the genetic relationship of cultivated and wild Atractylodes and its closely related species by using the Internal Transcribed Spacer 2(ITS2)barcode,in order to explore the cultivation origin of A. coreana from Northeast China. Method: Genomic DNAs were extracted from 40 samples of Atractylodes and its closely related species from different cultivated habitats, and 7 samples of wild A. coreana. were also extracted. The ITS2 sequences of these samples were amplified, and bidirectional sequencing was conducted by polymerase chain reaction(PCR). Totally 47 ITS2 sequences were aligned by using MEGA 5.0,5.8S and 28S sequences were removed to obtain the complete ITS2 sequence and build neighbor-joining (NJ) tree. Result: The lengths of ITS2 sequences of all samples were 232 bp. The NJ tree and the secondary structures of ITS2 showed that various varieties could be distinguished obviously except A. chinesis and A. coreana, which showed a good monophyly. The NJ tree showed that cultivated and wild A. coreana can also get together very well. Conclusion: As a DNA barcode,ITS2 sequences can be used to stably and accurately distinguish various varieties of Atractylodes. The relationship between A. chinesis and A. coreana is very close. A. coreana can be considered as a variant of A. coreana in North China. It is recommended to incorporate A. coreana into A. chinesis. The large-scale cultivation of A. coreana may originate from local wild population in Liaoning province, and the provenance may come from Xiuyan and other places in Liaoning province.

RESUMO

BACKGROUND: Intestinal and lymphoid tissues constitute an important part of intestinal immunity, which plays an important regulatory role in spleen deficiency and hydronephrosis. OBJECTIVE: To observe the effect of acupuncture on T lymphocyte subsets in lymph nodes of rats with spleen deficiency, and to investigate the correlation of spleen deficiency with intestinal immunity and the mechanism of acupuncture for spleen deficiency syndrome. METHODS: Thirty-six female Sprague-Dawley rats were randomly divided into three groups: model, acupuncture and blank control groups. The rat model of spleen deficiency was established by fatigue-induced spleen injury plus abnormal diet for 31 days. Afterwards, the rats in the acupuncture group received acupuncture at Zusanli(ST 36).Urine D-xylose excretion rate was detected during modeling and treatment. Then, the mesenteric lymph nodes were removed, and the changes in T lymphocyte subsets in the mesentericlymph nodes were observed by immunohistochemistry. RESULTS AND CONCLUSION: Urine D-xylose excretion rate under spleen deficiency in the modeling and acupuncture groups was significantly lower than that in the blank control group (P < 0.05 or P < 0.01); after acupuncture, the urine D-xylose excretion rate was significantly increased compared with the modeling group (P < 0.01), but still lower than that in the blank control group (P < 0.05). The count of CD4+T lymphocytes, count of CD8+T lymphocytes and ratio of CD4+/CD8+T lymphocytes were ranked as follows: blank control group >acupuncture group>modeling group(P<0.01 or P<0.05).These results suggest that acupuncture at Zusanli can improve the urine D-xylose excretion rate, regulate the balance of T lymphocyte subsets in mesenteric lymph node of rats with spleen deficiency, thus improving the intestinal immune function, spleen deficiency systems, disorder of intestinal digestive function, intestinal digestion and absorption, as well as anorexia, loose stool, diarrhea and other symptoms of the digestive system.

RESUMO

@#BACKGROUND: Triggering receptor expressed on myeloid cells-1 (TREM-1) is a cell surface receptor expressed on neutrophils and monocytes. TREM-1 acts to amplify inflammation and serves as a critical mediator of inflammatory response in the context of sepsis. To date, the predisposition of TREM-1 gene polymorphisms to septic shock has not been reported. This study was designed to investigate whether TREM-1 genomic variations are associated with the development of septic shock. METHODS: We genotyped two TREM-1 single nucleotide polymorphisms (SNPs, rs2234237 and rs2234246) and evaluated the relationships between these SNPs and septic shock on susceptibility and prognosis. RESULTS: TREM-1 rs2234246 A allele in the promoter region was significantly associated with the susceptibility of septic shock in recessive model (AA, OR=3.10, 95%Cl 1.15 to 8.32, P=0.02), and in codominant model (AG, OR=0.72, 95%Cl 0.43–1.19, P=0.02; AA, OR=2.71, 95%Cl 1.00–7.42;P=0.03). However, in three inherited models (dominant model, recessive model, and codominant model), none of the assayed loci was significantly associated with the prognosis of septic shock. The non-survivor group demonstrated higher plasma IL-6 levels (99.7±34.7 pg/mL vs. 61.2±26.5 pg/mL, P<0.01) than the survivor group. Plasma concentrations of IL-6 among the three genotypes of rs2234246 were AA 99.4±48.9 pg/mL, AG 85.4±43 pg/mL, and GG 65.3±30.7 pg/mL (P<0.01). The plasma concentrations of IL-6 in patients with AA genotypes were significantly higher than those in patients with GG genotypes (P<0.01). CONCLUSION: TREM-1 genetic polymorphisms rs2234246 may be significantly correlated only with susceptibility to septic shock in the Chinese Han population.

RESUMO

Objective To develop a blood gas,acid-base imbalance analysis computer software to diagnose systematically, rapidly, accurately and automatically determine acid-base imbalance type, and evaluate the clinical application. Methods Using VBA programming language,a computer aided diagnostic software for the judgment of acid-base balance was developed. The clinical data of 220 patients admitted to the Second Affiliated Hospital of Guangzhou Medical University were retrospectively analyzed. The arterial blood gas〔pH value,HCO3-,arterial partial pressure of carbon dioxide(PaCO2)〕and electrolytes included data(Na+and Cl-)were collected. Data were entered into the software for acid-base imbalances judgment. At the same time the data generation was calculated manually by H-H compensation formula for determining the type of acid-base imbalance. The consistency of judgment results from software and manual calculation was evaluated, and the judgment time of two methods was compared. Results The clinical diagnosis of the types of acid-base imbalance for the 220 patients:65 cases were normal,90 cases with simple type, mixed type in 41 cases, and triplex type in 24 cases. The accuracy of the judgment results of the normal and triplex types from computer software compared with which were calculated manually was 100%,the accuracy of the simple type judgment was 98.9% and 78.0% for the mixed type, and the total accuracy was 95.5%. The Kappa value of judgment result from software and manual judgment was 0.935,P=0.000. It was demonstrated that the consistency was very good. The time for software to determine acid-base imbalances was significantly shorter than the manual judgment(seconds:18.14±3.80 vs. 43.79±23.86,t=7.466,P=0.000),so the method of software was much faster than the manual method. Conclusion Software judgment can replace manual judgment with the characteristics of rapid, accurate and convenient, can improve work efficiency and quality of clinical doctors and has great clinical application promotion value.

RESUMO

Objective To explore the influence of occupational stress and negative life events that occur during pregnancy on different types of low birth weight (LBW). Methods 438 singleton LBW infants (birth weight of less than 2500 g and their pregnancy term from 28 to 42 weeks) were selected as case group, and they were further divided into symmetric LBW infants (337 cases) and asymmetric LBW infants (101 cases). According to situation of each LBW infant, a singleton with full term and normal birth weight was selected as control group matched by sex, pregnancy term, time during delivery and types of hospital. All of their mothers were inquired by well trained investigators on their socio-demographic characteristics, occupational stress, and negative life events that occurred in different pregnancy term. After controlling for mother's age, occupation, education level and family income, multinomial logistic regression was employed to asses the influence of occupational stress and negative life events on symmetric LBW and asymmetric LBW. Results Compared with those using low technical skills, mothers with high technical skill utilization significantly decreased the risk of laboring both symmetric LBW ( OR = 0.69, 95 % CI:0.49-0.98) and asymmetric LBW (OR = 0.53,95%CI: 0.31-0.89). Compared with those without exposure to negative life events, mothers with negative life event score ≥ 3 in the whole duration of pregnancy had significantly increased the risk of delivering symmetric LBW ( OR = 2.30, 95% CI : 1.08-4.88), mothers with negative life event score ≥3 in the middle three months of pregnancy, ≥3 in the last three months of pregnancy, = 2 and ≥3 in the whole duration of pregnancy had significantly increased the risk of delivering asymmetric LBW, and their OR (95 % CI ) was 8.85 (1.97-39.68), 3.80 ( 1.40-10.29 ),3.58(1.33-9.66) and 3.48 (1.32-9.13), respectively. Conclusion Occupational stress and negative life events might produce different influence on symmetric LBW and negative life events that occurr in the different terms of pregnancy had different impact on symmetric LBW and asymmetric LBW.
